Developed in the 1960s, this technology has become a key bridge connecting biocatalysis and industrial applications. Its core value lies in overcoming the limitations of natural enzymes in industrial use: free enzymes are difficult to recycle and reuse in reaction systems, have poor stability, and are easily inactivated by environmental factors. Immobilization technology, by combining enzymes with carriers, constructs a reusable, stability-enhanced heterogeneous catalytic system, significantly improving the economy and applicability of enzymes. Currently, the mainstream immobilization methods include adsorption, covalent binding, cross-linking, and encapsulation. Adsorption methods rely on weak interactions such as van der Waals forces and ionic bonds to immobilize enzymes on a support surface. While simple to operate, they are sensitive to pH and ionic strength. Covalent binding methods form strong covalent bonds between enzyme molecules and the support. While complex, they also offer low enzyme leakage. Crosslinking methods utilize bifunctional reagents to crosslink enzyme molecules, forming a network structure. Encapsulation methods confine enzymes within polymer gel grids or microcapsules, effectively preserving their spatial conformation. Each of these techniques has its own unique characteristics, and the optimal approach should be selected based on the enzyme's characteristics and application scenario.
The advantages of immobilization technology manifest in multiple dimensions: first, significantly improving operational stability, enhancing the enzyme's ability to withstand temperature and pH fluctuations, and organic solvent shock; second, enabling repeated batch use or continuous production of the enzyme, significantly reducing catalyst costs; third, easy separation of immobilized enzymes from the reaction system, ensuring product purity and preventing enzyme contamination; and even more importantly, some immobilized enzymes exhibit enhanced properties, such as altered substrate specificity or improved reaction selectivity. However, this technology also faces challenges, such as potential loss of enzyme activity during the immobilization process, increased production costs for support materials, and reduced reaction rates due to mass transfer resistance. In practical applications, immobilization technology has profoundly transformed biomanufacturing. Immobilized glucose isomerase is used in fructose-glucose syrup production, enabling continuous sugar conversion. Immobilized penicillin acylase plays a key role in antibiotic semi-synthesis. Immobilized enzymes are also actively used in chiral drug synthesis, biosensor construction, and environmental pollutant degradation. With the development of new carrier materials such as nanomaterials and smart hydrogels, as well as new strategies such as targeted immobilization and multi-enzyme co-immobilization, enzyme immobilization technology is evolving toward higher efficiency, greater stability, and more precise control, continuously injecting innovative momentum into green manufacturing and sustainable development.
