Biodegradable magnesium alloys as a new generation of biomedical materials have many advantages over traditional materials, in line with the development trend of biomedical materials. It can be used for the development of unique new medical devices. Its clinical applications will have a milestone in the development of biomedical materials.
Since the beginning of this century, a new generation of biodegradable medical metal materials represented by magnesium alloys has attracted people"s attention and been extensively studied. Such materials not only inherit the good mechanical properties and processing properties of metal materials, but also can be degraded in the human body. Due to its good biocompatibility, mechanical properties and degradable properties, magnesium alloys are known as revolutionary medical metal materials. Compared with traditional bone repair materials, magnesium alloys have the following advantages: (1) Degradability. Mg has a low standard electrode potential (-2.37 V), and is easily corroded in corrosive media. As an intraosseous implant material, this feature allows it to degrade in the human body after completing bone repair or fixation functions, thereby avoiding secondary operations and greatly reducing the risk and burden of patients. (2) Good biomechanical compatibility. Compared with traditional metal bone repair materials, the physical and mechanical properties of magnesium alloys are closer to those of bone tissue, which can effectively avoid the stress shielding effect. The density of magnesium alloy is 1.74~2.00 g/cm3, which is close to that of cortical bone. The elastic modulus of magnesium alloy is 41-45 GPa, which is slightly higher than that of cortical bone and much lower than that of stainless steel and titanium alloy. At the same time, as a metal material, magnesium alloy has high strength and toughness, the tensile strength is 4-15 times that of polylactic acid (PLA), and the fracture toughness is much higher than that of hydroxyapatite (HA). Therefore, magnesium alloys have high mechanical strength and good mechanical compatibility. (3) Good biocompatibility. Mg is one of the essential nutrients for the human body, and its content in cations in the body is second only to Ca, K, and Na. The normal Mg content in an adult is 21–28 g, of which about 53% exists in bone, 27% in muscle tissue, 19% in soft tissue, and a small amount of the rest exists in blood, liver, brain, kidney and other organs. Mg can catalyze and activate more than 300 kinds of enzymes. It is an important activator of various enzymes in cell metabolism and participates in various physiological activities such as protein and DNA synthesis, energy storage and transportation, nerve signal conduction, and muscle contraction. Lack of Mg may cause arrhythmia, hypertension, ischemic heart disease, cerebral infarction, osteoporosis and other diseases. The World Health Organization recommends a daily intake of Mg of 280-300 mg for adults and 250 mg for children. Therefore, magnesium alloy has a good biosafety basis as a biodegradable medical material. (4) Good osteoinductivity. A large number of studies have shown that magnesium ions can induce new bone formation.
