In the MST experiment, the sample is heated by an infrared laser to generate a microscopic temperature gradient field, and then the directional movement of molecules is monitored and quantified by covalently bound fluorescent dyes, fluorescent fusion proteins or autofluorescence. This movement reflects the thermophoresis behavior of molecules in the temperature gradient field, and the interaction between molecules can be inferred.
The MST experimental process is relatively simple and does not require complex sample processing steps. From the start of the experiment to data analysis, the entire process can be completed in a short time, usually within 20 minutes. MST technology can detect biomolecular interactions ranging from pM to mM levels and has a wide range of applicability. MST technology is not only suitable for the study of interactions between small molecules, proteins, nucleic acids and other biomolecules, but can also be extended to a wider range of fields such as ions and ribosomes. MST experiments can be performed in a variety of buffers, including complex systems such as serum and cell lysates.
