Single-use plastic systems, the types used for drug delivery and manufacturing, can vary in size from 1 liter to 2,500 liters and although they can cost in the thousands of dollars they are still considered cheaper than the traditional cleaning route for the steel reactors.
A typical single-use bioreactor is a plastic bag made of a multilayered polymer film. The three main layers are the inside layer, which is in contact with the cell culture; a barrier layer, which prevents gas diffusion into and out of the bag; and an outer layer, which provides mechanical stability. Those layers are “glued” together with tie layers that bridge their disparate chemical properties.
In the North America market, the major players of disposable bags for bioreactors include Entegris, Corning, Sartorius, Thermo Fisher Scientific and Danaher, and the top five players account for about 75% of the market share. In terms of product types, three-dimensional bags were the largest market segment, accounting for 59 percent of the market share, while in terms of applications, MAB and recombinant proteins accounted for the largest share at about 48 percent.
At present, the global biopharmaceutical single-use system (SUS) market is mainly occupied by overseas giants such as Danaher, Thermo Fisher, Sartorius and Merck. Dongfulong, Chutian Technology, Duoning Bio, Lechun Bio, Jinyi Shengshi and others have made early layouts, but their advantages are still limited to related peripheral consumables.
In recent years, single-use reaction technology has gradually become the choice of biopharmaceutical companies and CDMO companies. Due to its low pollution risk, cleanliness and low verification cost, it has been well received in the industry. The choice of single-use technology not only brings an innovative direction for biopharmaceutical companies and CDMO companies, but more importantly, in the production process, it greatly shortens the construction and production time, reduces the floor space, and shortens the waiting time for drug research and development.
At present, single-use bioreactors are widely used for seed expansion culture, including small to pilot-scale mammalian cells, plant cells and insect cells. It is the mainstream production process for monoclonal antibodies, vaccines, recombinant proteins, etc. For single-use bioreactors, the limitation of reactor scale and the problems that may be caused by membrane extracts and precipitates have always been a long-term problem for the industry.
Specifically, on the one hand, disposable materials are difficult to withstand continuous high pressure, which limits the further expansion of the scale of disposable bioreactors. On the other hand, disposable plastic bags used for cell culture may release dissolution when in contact with the culture medium. Once the latter enters the culture medium, it will hinder the growth of cells. At present, there is no standard test method that can be applied to the detection of dissolutions of different membranes. The optimization of these underlying technologies is undoubtedly a process of long-term experience accumulation and repeated trial and error.
