Spermidine is a naturally occurring polyamine compound and one of the important organic molecules in living organisms. It plays an important role in cell growth, metabolism and survival. Spermidine mainly exists in the cytoplasm and nucleus. It is a polyamine with the chemical formula (NH2(CH2)3NH(CH2)4NH2).
Spermidine raw materials are sourced via two routes: "natural extraction" and "synthetic/fermentation." Natural extraction primarily uses wheat germ, buckwheat sprouts, or Chlorella powder, containing 0.1–0.5% spermidine. After water extraction, decolorization, ion exchange, and concentration/crystallization, the raw material cost accounts for approximately 30% of the ex-factory price, but the yield is low and waste is significant. The synthetic route centers on acrylonitrile-putrescine Michael addition-high-pressure hydrogenation. Putrescine (butanediamine) and acrylonitrile together account for 55% of the raw material cost, while hydrogenation using Raney nickel, solvent recovery, and purification processes account for 25%. Currently, this remains the most economical process for kilogram-scale production.
Although fermentation uses glucose and inorganic nitrogen as a cheap carbon source, the product concentration is ≤ 3 g L⁻¹, and the costs of downstream concentration, decolorization, and impurity removal drive up the overall cost, making it 15–20% higher than chemical methods. It has not yet been scaled up to the ton level.
