Due to the lack of Rep gene, the recombinant AAV viral genome does not integrate into the host genome and exists as an extrachromosomal free body for a long time. These characteristics make AAV viral vectors a gene delivery tool with great application prospects. Compared with other viral vectors, AAV has many advantages: 1) Strong diffusibility: rAAV is small in size and high in titer. Compared with adenovirus and lentivirus, it has higher diffusivity and can cross the blood-brain barrier, making it an ideal tool for infecting the nervous system; 2) High specificity: rAAV has multiple serotypes, and different serotypes have different recognition and infection abilities for different tissues. It is a tool virus preferred by animal experiments; 3) High safety: rAAV persists outside the chromosome, reducing the risk of mutation, and has low immunogenicity, so it can be directly used for human gene therapy.
